Phenotypic switch of smooth muscle cells in paediatric chronic intestinal pseudo‐obstruction syndrome

چکیده
Smooth Muscle Cells (SMC) are unique amongst all muscle cells in their capacity to modulate phenotype. Indeed, SMCs do not terminally differentiate but instead harbour a remarkable dedifferentiate, switching between quiescent contractile state and highly proliferative migratory phenotype, quality often associated SMC dysfunction. However, phenotypic plasticity remains poorly examined the field of gastroenterology particular pathologies which gut motor activity is impaired. Here, we assessed status biopsies infants with chronic intestinal pseudo-obstruction (CIPO) syndrome, life-threatening motility disorder. We showed that CIPO-SMCs decreased level markers. This phenotype accompanied by an increase Platelet-Derived Growth Factor Receptor-alpha (PDGFRA) expression. this modulation occurs without origin-related differences CIPO circular longitudinal-derived SMCs. As characterized PDGFRA as marker digestive mesenchymal progenitors during embryogenesis, our results suggest switch CIPO-SMC towards undifferentiated stage. The development culture characterization should enable us design therapeutic approaches promote differentiation CIPO.

C ardiovascular diseases, such as ischemic heart disease and stroke, represent the number one cause of death worldwide. 1 In most cases, atherosclerosis and hypertension constitute the underlying causes for cardiovascular disease because it leads to arterial occlusion and impaired cardiac function, respectively. متن کاملPhenotypic Modulation of Vascular Smooth Muscle Cells:
G protein-coupled receptors regulate two major functions of vascular smooth muscle cells (SMCs) the vasomotor contractile response and the proliferative response. The contractile phenotype of SMCs has mainly been studied in situ within intact blood vessels, whereas the synthetic or proliferative SMC phenotypes can be studied in culture and represent a model of the SMC phenotype seen in vascular...
متن کاملAn RGS4-Mediated Phenotypic Switch of Bronchial Smooth Muscle Cells Promotes Fixed Airway Obstruction in Asthma
In severe asthma, bronchodilator- and steroid-insensitive airflow obstruction develops through unknown mechanisms characterized by increased lung airway smooth muscle (ASM) mass and stiffness. We explored the role of a Regulator of G-protein Signaling protein (RGS4) in the ASM hyperplasia and reduced contractile capacity characteristic of advanced asthma. Using immunocytochemical staining, ASM ...

Chronic idiopathic intestinal pseudoobstruction is a serious disorder of intestinal neuromuscular function resulting in recurrent episodes of intestinal obstruction, and is caused by primary disease of the enteric nerves or enteric smooth muscle. Gastric electrical control activity detected by the non-invasive technique of surface electrogastrography was investigated in 11 children (0.1-16 year...
متن کاملPhenotypic alteration of vascular smooth muscle cells precedes elastolysis in a mouse model of Marfan syndrome.
Marfan syndrome is associated with early death due to aortic aneurysm. The condition is caused by mutations in the gene (FBN1) encoding fibrillin-1, a major constituent of extracellular microfibrils. Prior observations suggested that a deficiency of microfibrils causes failure of elastic fiber assembly during late fetal development. Mice homozygous for a targeted hypomorphic allele (mgR) of Fbn...
